核酸适体筛选方法的研究进展

摘要(Abstract):

核酸适体是能以极高的亲和力和特异性与靶分子结合的寡核苷酸序列,可识别的靶分子范围非常广,理论上自然界存在的任何物质均可通过指数富集的配体系统进化(systematic evolution of ligands by exponential enrichment,SELEX)技术,筛选到其相应的核酸适体。近年来,在传统SELEX技术的基础上,新的SELEX技术不断出现,如毛细管电泳SELEX(capillary electrophoresis SELEX,CE-SELEX)、荧光磁珠SELEX(Flu Mag SELEX)、自动化SELEX(automated SELEX)、细胞-SELEX(cell-SELEX)、消减SELEX(subtractive SELEX)技术等。这些新技术的出现,在不断完善SELEX技术的同时,也极大地推动了适体技术在各学科领域中的发展及应用。本文对核酸适体的筛选方法进行了总结和展望,为核酸适体的筛选提供了参考。
Aptamers are oligonucleotides that can bind to a wide range of target molecules with high affinity and specificity. The aptamers to all target molecules in nature may be theoretically screened by systematic evolution of ligands by exponential enrichment(SELEX). In recent years, on the basis of traditional SELEX technology, new SELEX technologies are emerging such as capillary electrophoresis SELEX(CE-SELEX), fluorescent magnetic beads SELEX(Flu Mag SELEX),automated SELEX and cell-SELEX, subtractive SELEX. The new technologies improved the SELEX process gradually and promoted their application in various fields. This paper summarized and prospected the methods for screening of aptamers and provided a reference for aptamers screening.
