摘要(Abstract)：

目的探讨雪胆甲素(CucurbitacinⅡa)对人非小细胞肺癌(Non-small cell lung cancer,NSCLC)A549细胞增殖的影响。方法以不同浓度的雪胆甲素作用于A549细胞和小鼠原代培养的脾细胞,采用MTT法检测雪胆甲素对两种细胞增殖的影响;流式细胞术及Annexin V-PE细胞凋亡试剂盒检测雪胆甲素对A549细胞细胞周期及凋亡的影响。结果雪胆甲素对A549细胞和小鼠原代脾细胞的增殖均有抑制作用,且呈浓度依赖性,25～100μmol/L的雪胆甲素对A549细胞的增殖抑制率明显高于脾细胞;雪胆甲素可将大部分A549细胞阻滞于G0/G1和S期,少部分细胞阻于G2和M期,并可促进细胞凋亡。结论雪胆甲素可明显抑制A549细胞增殖,在NSCLC高效低毒的治疗药物的开发中具有重要价值。
